Researchers at the University of Texas at Galveston have found that the enzyme aldose reductase is a critical link in the cascade of steps resulting in an allergen-provoked asthma attack, and that aldose reductase inhibitors, already in use for the treatment of diabetes, can prevent an asthma attack. Oral administration of aldose reductase inhibitors worked effectively to block inflammation in a series of in vitro experiments with ragweed extracts and human airway epithelial cells. This was followed by an experiment with asthmatic mice that were fed aldose reductase inhibitor before inhaling ragweed extract and the therapy reduced asthma. Further research could lead to a much needed alternative to the current asthma therapy involving corticosteroids and bronchodilators, which have many side effects. The group is moving on to clinical trials in humans. The study is published in the Aug. 6th edition of PLoS ONE.
Missing Link in Celiac Disease Identified
Researchers at the University of Maryland have identified a formerly mysterious protein called zonulin as the precursor to haptoglobin 2, a molecule that is responsible for gut permeability. “This molecule could be a critical missing piece of the puzzle to lead to a treatment for celiac disease, other autoimmune disorders and allergies and even cancer, all of which are related to an exaggerated production of zonulin/pre-haptoglobin 2 and to the loss of the protective barrier of cells lining the gut and other areas of the body, like the blood brain barrier,” says Dr. Fasano who headed the team’s study. The molecule is present in humans but not other primates and it is believed to have mutated from haptoglobin 1 some two million years ago. This may explain why apes rarely have autoimmune diseases, but humans currently suffer from 70 different kinds. Piers of Dr. Fasano’s team have heralded this as “the molecule of the century”, and it may will lead to exciting breakthroughs in allergy and celiac disease. Dr. Fasano reported the discovery in the Sept. 7th online edition of the Proceedings of the National Academy of Sciences. (Thanks to Jen for pointing this out.)

Children’s Memorial Food Allergy Study
The Children’s Memorial Food Allergy Study in Chicago is still looking for families to participate in this long term family-based study of biological parents and at least one food allergic child (age 0-21 years). The goal is to answer fundamental questions that have never been systematically studied on a large scale: What is the cause of food allergy? How can food allergy be predicted and prevented? Are there alternative or better treatments for food allergy? This is the largest genetic study of food allergy ever and the answers to these basic questions will point toward their goal, which is nothing less that a cure for food allergy.
Each family member that participates receives a $25 gift card to Target, free parking, additional gift card(s) to cover travel expenses and free blood RAST screening for allergies. The test alone can cost several hundreds of dollars, and later you can tell your everyone that you were part of the solution to the need for gluten free foods. Since inception the study has enrolled 1000 families. An additional 500 families are needed in order to secure additional NIH funding for this single largest genetic study of food allergy.
